PURPOSE:To separate pure cholesterol oxidase, easily, from an enzyme solution containing cholesterol oxidase, by the use of cholesterol as adsorbent. CONSTITUTION:Floating cholesterol (floating on water surface) is converted to precipitated cholesterol by heating. An enzymatic solution containing cholesterol oxidase (medium obtained by culturing cholesterol oxidase-producing microbials) is treated with the above precipitated cholesterol, and adsorbed thereto. The adsorbed cholesterol oxidase is washed, eluted with a surface active agent, and purified to afford the pure cholesterol oxidase. |
