PURPOSE:To prepare high purity plasmid useful as a vector for the in vitro gene recombination, by the density gradient centrifugal separation of Brevibacterium lactofermentum which is an of amino acid fermentation bacterium. CONSTITUTION:Bacterial cells of Brevibacterium lactofermentum ATCC13869, which is an amino acid fermentation bacterium, are mildly disintegrated, and the cytoplasm DNA is selectively extracted. It is centrifuged in cesium chloride/ethidium bromide density gradient to separate the plasmid having a molecular weight of about 3.7X10<7>. The density of cesium chloride is adjusted to 1.57-1.58g/ml, pref. to 1.575g/ml. |
