<p>Purification and concentration of influenza virus containing practically no viral protein or other extraneous impurity. The method gives a pure form of virus. The culture medium containing the influenza virus is centrifuged, re-suspended and absorbed by addition of 10 to 15% barium sulphate, the barium sulphate suspension of the virus is washed with 0.1 to 0.5% sodium or potassium citrate in a 0.5 to 2% solution of sodium chloride. The virus is eluted from the barium sulphate by a solution of 7.5 to 15% sodium or potassium citrate and purified by filtering the eluate over a layer of diatomaceous earth and then filtering through one or more membrane filters.</p> |
