<p>PURPOSE:To obtain a number of tobacco seedlings in a short period by inoculating pieces of the organs of a Nicotiana tabacum with Agrobacterium rhizogenes MAFF-03-01724 followed by making a culture to directly develop many shoots from the infected sites. CONSTITUTION:Pieces of the organs such as leaves, stems or roots of a Nicotiana tabacum are inoculated with Agrobacterium rhizogenes MAFF-03-01724 followed by making a culture. In this case, for said tobacco plant, surface sterilization is made and said bacteria is then inoculated; alternatively, a sterilized plant made in advance may be used. Said culture is made in a solid medium e.g. at 25-28 deg.C under an illumination of ca.500-3000 lux. The shoots thus developed are cut and put to culture in a fresh medium to effect root development. The root development will occur in ca. 1-2 weeks, the resultant roots being then put to culture, thus completely obtaining tobacco- regenerated individual pieces within, in general, 1.5 months.</p> |
