IMMUNOLOGICAL TEST SUBSTANCE FOR RUBELLA AND PRODUCTION AND USE THEREOF

To prepare the new soluble rubella antigen, (a) the growth media for a tissue culture of cells infected with the rubella virus are subjected to a chromatographic separation on a column consisting of a solid phase to which has been attached IgG derived from human serum known to contain antibodies reactive against rubella antigens; (b) this column is eluted in order to separate the substances attached to the IgG which it contains; (c) the separated eluted substance is subjected to permeation gel chromatography in order to separate the substance on the basis of the molecular size; (d) the rubella antigen is isolated from the said separated eluted substance classified on the basis of the relative reversed passive haemagglutination activity. The antigen thus obtained can be used to prepare immunological reagents composed of particulate materials whose surface is sensitised by the antigen and which are used for the detection of rubella antibodies in direct agglutination tests.
