VERFAHREN ZUR ELEKTROPHORETISCHEN UNTERSUCHUNG VON PROTEINEN

In a new procedure for the electrophoretic examination of proteins (enzymes, serum proteins, tissue, homogenates) involving introduction of the proteins by means of a carrier medium into holes in a flat layer of carrier gel (agar, starch or polyacrylamide gel), there is used as the carrier medium a swollen gel (preF. with a low degree of crosslinking) in the form of dextran swollen with water or buffer soln. with the addition of sucrose. Use of sucrose-contg. dextran gel as carrier medium enables protein to be applied to the carrie medium by simple pipetting. the sucrose stabilises the proteins against damage while passing through the dextran. Larger amts. of protein can be used than with known methods, and even minor proteins (e.g. human erythrocyte phospho-glucomutase isoenzyme 3) can be detected.
