1460471 Benzopyrano[4,3-c]pyridines ABBOTT LABORATORIES 12 July 1974 [13 July 1973 8 April 1974] 31075/74 Heading C2C The invention comprises compounds of formula and their pharmaceutically acceptable salts, wherein R 1 is C 1-6 alkyl; R 2 is C 1-6 alkyl substituted by C 3-7 cycloalkyl, or C 1-20 alkyl, or Ph-C 1-10 -alkyl (in which Ph may contain up to 3 of halogen, CF 3 and C 1-6 alkyl substituents); R 3 is OH, acyloxy, up to C 6 alkoxy, alkenyloxy or alkynyloxy, or R 7 R 8 N-C 1-8 alkyl-COO, where R 7 and R 8 are each H or C 1-6 alkyl, or NR 7 R 8 forms a saturated heterocyclic group (as defined in the Specification). The compounds where R 3 = OH are prepared by dehydrogenation of the 1,2,3,4-tetrahydro-analogue (with simultaneous hydrogenolysis of a 2-benzyl group if present), and may then be etherified or esterified. Therapeutic compositions having analgesic, anti-anxiety and anti-depressant activity comprise compounds of the above formula, and may be administered orally, parenterally or rectally. |
