Acetylated cardiotonic glucosides are produced by reacting the glucoside with an alkyl orthoacetate and submitting the resulting cyclic orthoacetate to partial saponification. The intermediate cyclic orthoacetate may be isolated and/or other free OH gps. acetylated if desired. This process enables acetyl gps. to be introduced into desired positions on the pyrannoside radical of the glocoside. Preferred products are alpha-digoxin, alpha-digitoxin, and 2'-acetyl-proscillaridin. The products are of value in the treatment of cardiac insufficiency. They are more readily absorbed than the parent glucosides.
|
