1504442 Naphthalene derivatives BEECHAM GROUP Ltd 25 Nov 1976 [21 Jan 1976] 02275/76 Heading C2C Novel compounds I in which A is -CH(R 1 )-CH 2 - or where R 1 is H or CH 3 , and B is -CO- or -CH(OH)-, are prepared by (a) reacting a compound II in which X is Cl or Br, with a methyl metal compound and thereafter, if desired, reducing the -CO- group to a -CH(OH)- group, or (b) for compounds I in which B is -CO-, by reacting a compound III in which Q is -CH 2 - or -CH(CH 3 )-, with a base under anhydrous conditions, or (c) for compounds I in which A is -CH=CH-, by reacting 6 - methyl - 2 - naphthaldehyde with acetone in the presence of a base or with a Wittig reagent of formula Q 1 =CH-B-CH 3 where Q is a trisubstituted phosphorus atom. 2 - Bromomethyl - 6 - methylnaphthalene is prepared by reacting 2,6-dimethylnaphthalene with bromine. Pharmaceutical compositions having antiinflammatory activity for oral, parenteral, topical or rectal administration, comprise a compound I together with a pharmaceutical carrier. |
