1414848 Isolating biologically active materials CESKOSLOVENSKA AKADEMIE VED 2 May 1973 [10 May 1972] 20889/73 Heading A5B [Also in Divisions B1 and C3] In a method for the isolation of a soluble biologically active compound (B.I.C.) capable of forming a sorption complex with another B.I.C., by affinity chromatography based on the formation of the sorption complex between the B.I.C. desired to be isolated and the other B.I.C. linked to a solid carrier by a covalent bond, the carrier is a hydrophilic macroporous co-polymer of a hydrophilic monomer with a divinyl or polyvinyl monomer, and the B.I.C. is an enzyme, an enzyme inhibitor, an antibody, an antigen or a hormone. All the polymers disclosed are acrylic. The carrier may be a layer or column or may be in the form of a block, film, rod, string, rope, tape or belt. In examples, the pairs of B.I.C's are (1) chymotrypsin and trypsin-inhibitor; (2) trypsin and trypsin-inhibitor; (3) insulin (an antigen) and its antibody; (4) papain SH-proteinase and p-aminophenyl mercury acetate (an inhibitor); (5) chymotrypsin-inhibitor and chymotrypsin; (6) hen ovoinhibitor and trypsin; and (7) trypsin inhibitor and trypsin.
