<p>1466924 4 - Amino - indazole derivatives WANDER Ltd 2 April 1974 [5 April 1973] 14646/74 Heading C2C The invention comprises novel compounds of the Formulµ I and II wherein R 1 is H or C 1-3 alkyl, R 2 is H, halogen, OH or C 1-3 alkyl, alkoxy or alkylthio, R 3 is H, halogen, OH or C 1-3 alkyl or alkoxy, A is dimethylene or trimethylene wherein one hydrogen may be replaced by OH or C 1-3 alkyl and R 4 is -YR 5 , -NHNO 2 or NHR 5 where Y is oxygen or sulphur and R 5 is hydrogen or C 1-3 alkyl and acid addition salts thereof. Compounds of the Formula I may be prepared by (a) reacting a compound of the Formula II with a compound NH 2 -A-NH 2 (III), (b) cyclizing a compound of the Formula V or (c) reacting a compound of the Formula VI with a compound III. Compounds of the Formula II may be prepared by (a<SP>1</SP>) saponifying a compound of the Formula VIII to produce II wherein R 4 is OH or mercapto, (b<SP>1</SP>) alkylating a product of process (a<SP>1</SP>) to produce II where R 4 is C 1-3 alkylthio or alkoxy, (c<SP>1</SP>) reacting a compound of the Formula IV with cyanamide or a C 1-3 alkylcyanamide in the presence of an acid to produce II in which R 4 is NH 2 or NHC 1-3 alkyl, or (d<SP>1</SP>) reacting a compound of the Formula IV with methylnitrosonitro-guanidine to produce a compound II in which R 4 is NHNO 2 . N - (2 - Aminoethyl) - N<SP>1</SP> - (3 - chloro - 1H- indazolyl-4)thiourea is prepared by reduction of 3 - chloro - 4 - nitro - 1H - indazole to 4-amino- 3-chloro-1H-indazole followed by reaction with thiophosgene to produce 3-chloro-4-isothiocyan- 1H-indazole and then reaction with ethylene diamine. 4 - Cyanamido - 1H - indazole is prepared by dehydrosulphurization of (1H-indazolyl-4-)thiourea. Pharmaceutical compositions of a compound I or II or a pharmaceutically acceptable acid addition salt thereof show antihypertensive action when administered as a tablet or solution.</p> |
