PURPOSE:To obtain the beta-mannanase useful for obtaining mannose, galactose, glucose, mannooligosaccharide, etc., from 1'-Dmannane, easy in production and purification, and excellent in stability. CONSTITUTION:The beta-mannanase is obtained from the culture solution of a microorganism AM-001 strain (FERM P-8856) belonging to the genus Bacillus having the optimal growth pH in an alkaline range, etc., and has following physicochemical properties: (1) specifically acts on on beta-mannane, does not act on alphamannane, and acts on and hydrolyzes mannooligosacchrides having larger mol. wts. than that of beta-1,4-D-mannotetraose; (2) has the optimal pH of 8-10 and is stable at a pH of 6-10 under conditions of 60 deg.CX30min; (3) has the optimal temperature at approximately 60 deg.C, is stable at temperatures up to 65 deg.C under a condition of pH 8.0X30 deg.C, and is perfectly inactivated at 80 deg.C; (4) has an isoelectric point of 5.0-5.4 (by a chromato-focusing method); and (5)) has mol. wts. of 43000+ or -3000 and 57000+ or -3000 (SOS-polyacrylamide electrophoresis method). |
