1492829 D-amino acids or peptides bound to polymers CESKOSLOVENSKA AKADEMIE VED 22 April 1975 [25 April 1974] 16609/75 Headings C3P and C3H A sorbent comprises a D-amino acid or a peptide containing D-amino acid units, which may be optionally protected bonded to a hydrophilic polymeric carrier formed from a copolymer of hydroxyalkyl esters or hydroxyalkylamides of acrylic or methacrylic acid crosslinked with acrylate or methacrylate comonomers. Prior to condensation of the D-amino acid, the copolymer may be reacted with diamines (preferably of 1 to 10 carbon atoms) amino acids (preferably of up to 6 carbon atoms or dicarboxylic acids (preferably of up to 6 carbon atoms). The material may be synthesized on the amino modified carrier by reaction of amino-protective amino acid with the carrier, removal of the amino protective group and further condensation(s) and deprotection(s) with other N-protected amino acids. In the table, Z-Gly-D-Leu-OH, Z-Gly- D-Phe-OH, Z-Gly-D-Phe-L-Leu-OH, Z-Gly-LPhe - L - Leu - Gly - L - Phe - D - Leu - OH, Ac - DL - Lys - (Z) - OH, Z - D - Leu - OH, and Nps-D-Leu-OH are reacted with an amino modified gel of polyhydroxyalkylmethacrylate; and H-D-Phe-OCH(C 6 H 6 ) 2 , is reacted with a carboxyl modified gel of polyhydroxyalkylmethacrylate. In the examples; 2-hydroxyethylmethacrylate/ethylene dimethacrylate copolymer modified with 1,6-hexamethylenediamine is reacted with (1) Z-Gly-D-Leu-OH, (3) Boc-Leu- OH, Boc-D-Phe-ODcha, Z-Gly-OH (Boc groups removed after each condensation) and Z-Gly-OH (4) Z-D-Leu-OH, and Z group removed; (2) 2- hydroxyethylmethacrylate/ethylene dimethacrylate copolymer modified with #-aminocaproic acid reacted with H-D-Phe-OH; (5) 2-hydroxyethylacrylate/ethylene diacrylate modified with 1,6-hexamethylenediamine reacted with Z-Gly- D-Phe-OH; (6) 2-hydroxypropylmethacrylamide/ethylene dimethacrylate modified with 1,6-hexamethylenediamine reacted with Z-Gly- D-Leu-OH; (7) 2-(2-hydroxyethyl)hexylmethacrylate/1,11-trimethylolpropane trimethacrylate modified with ethylene diamine reacted with Z-Gly-D-Phe-L-leucine; (8) tetraethyleneglycol monomethacrylate/2,2<SP>1</SP> - dimethylpropanedioldimethacrylate modified with 1,10-diaminodecane reacted with H-D-Phe-OH. The products are used in affinity chromatography to purify (1), (5), (6), (8) chymotrypsin; (2) carboxypeptidase A; (4) aminopeptidase from a proteolytic enzyme preparation derived from Aspergillus flavus; (7) trypsin. |
