1491816 Making urokinase ABBOTT LABORATORIES 6 Oct 1975 [31 March 1975] 40878/75 Heading C3H Urokinase is produced from a contiguous culture of live kidney cells in an aqueous nutrient medium by addition to the usual cell culture maintenance additives of from 0À3 to 1-2 parts by weight of glycine for 100 parts by volume of the nutrient medium. Examples describe the pretreatment of human kidney cells and their treatment with a medium consisting of 0À5% by weight of lactalbumin hydrolysate, varying amounts of human serum albumin, 0À1% of glucose in Earle's salt, varying amount of glycine and 0À8 gm. per litre of NaHCO 3 . Titration is used to determine yield of urokinase. The culture temperature is preferably 37Œ 0À5‹ C.
