New L-Leucyl-p-aminoanilide derivs. are of formula (I): (where R1 and R2 are H or lower alkyl op. substd. by OH or MeSO2NH; R3 is H or lower alkyl). (I) are esp. useful as substrates for measuring the activity of leucylaminopeptidase in serum and tissue samples for diagnostic purposes. The analysis results obtd. with (I) are in good agreement with those obtd. using L-leucyl-beta-napthhylamide, and the formation of carcinogenic beta-naphthylamine is avoided. (I) is e.g. L-leucyl-4-diethylaminoanilide, prepd. by reacting t-butyoxycarbonyl-L-leucine with N, N-diethyl-p-phenylenediamine in the presence of DCC.
