Novel substituted benzamides of the formula wherein A and R are allyl or C 1-5 alkyl, X is C 1-5 alkyl or OH, Y is halogen, m and n are 1 to 3 and acid addition and quaternary ammonium salts thereof are prepared by reacting the appropriate benzoic acid chlorides or esters with the corresponding heterocyclic amines. 2 - Methoxy - 4 - methyl - 5 - chlorobenzoyl chloride is obtained by reacting thionyl chloride with 2 - methoxy - 4 - methyl - 5 - chlorobenzoic acid, obtained by chlorinating and hydrolysing 2 - methoxy - 4 - methyl - 5 - chloroacetophenone, resulting from methylating 2 - hydroxy - 4 - methyl - 5 - chlorotoluene, which is prepared by treating 2 - chloro - 5 - acetoxytoluene with aluminium chloride. Methyl - 2 - methoxy - 4 - hydroxy - 5 - chlorobenzoate is made by chlorinating methyl - 2- methoxy - 4 - hydroxybenzoate, obtained by the hydrogenolysis of methyl - 2 - methoxy - 4 - benzyloxybenzoate, resulting from the methylation of methyl - 2 - hydroxy - 4 - benzyloxybenzoate, which is prepared by reacting methyl 2,4-dihydroxybenzoate with benzyl chloride. Pharmaceutical compositions, having antiemetic, spasmogenic and analgesic activity, contain the above novel compounds or pharmacologically acceptable acid addition or quaternary ammonium salts thereof and carriers. The compositions may be in the form of tablets, phials, suppositories, granules, syrups, sugar coat pills, solutions or aerosols. |
