New anhydrofuranose derivs are of formula (I):- In (I) R2 and R3 are H, alkyl, alkenyl aralkyl or acyl and A-O is CHOR5-CH2O or CH(CH2OR6)O, where R5 and R6 are as defined for R2, or 2 of R2, R3 and R5 or R6 can form a lower alkylidene or aralkylidene gp.; with the proviso that (i) not all of R2, R3 and Rk or R6 are H; (ii) when A-O is CHOr5-CH2O, not all of R2, R3 and R5 are methyl, acetyl or tosyl; (iii) when A-O is CH(CH2OR6), not all of R2, R3 and R6 are benzyl. Cpds. (I) have fibrinolytic and thrombolytic activity. Those where R2, R3 and/or R5 or R6 are acyl residues of an anti-inflammatory carboxylic acid (esp. 1-/4-(1-cyclohexenyl) phenyl/propionyl) have anti-inflammatory and analgesic activity. A typical cpd. (I) is 1,5-anhydro-2-0- allyl-3,6-di-0-benzyl-alpha-D-galactofuranose. In an example, this is prepd. from 1-0-acetyl-2-0-allyl-3, 6-di-0-benzyl-4-0-mesyl-alpha-D-glucopyranose.
|
