PURPOSE:To exactly measure a mol. wt. by using the S-alkyl protein formed by alkylating the thiol groups formed by a reduction treatment. CONSTITUTION:The mol. wt. marker consisting of the S-alkyl protein formed by alkylating the thiol groups of the protein of a known mol. wt. having the thiol groups formed by the reduction treatment is used. The protein of a known mol. wt. refined by the conventional method is dissolved in a buffer soln. and a reducing agent is added thereto to effect a heating reaction and to cleave the disulfide bond of the protein. An alkylating agent is then added thereto and the reaction is effected for several hours preferably at room temp. or below to alkylate the thiol groups formed by the cleavage. The recombination of the thiol groups to each other is prevented in this way and the mol. wt. is exactly measured without generating side-product bands and tailing. |
