1496866 4,6,6 - Trimethyl - 2,4,5,6 - tetrahydrocyclopentapyrrole - 4 - carboxamides STERLING DRUG Inc 2 March 1976 [17 March 1975] 08385/76 Heading C2C Novel 4,6,6 - trimethyl - 2,4,5,6 - tetrahydrocyclopentapyrrole - 4 - carboxamide derivatives having the general formula wherein one of R 1 and R 3 is hydrogen or alkyl and the other is either (a) a group of the formula CH 2 OR 4 , where R 4 is carboxyalkanoyl or the ammonium salt thereof, alkanoyl, benzoyl or alkyl-substituted benzoyl, or (b) a group of the formula wherein R 5 is hydrogen or alkyl; R 6 is phenyl, alkenyl, cyano, alkyl, alkynyl, carboalkoxyalkylene, or carboalkoxyalkynyl; and R 2 is alkyl or phenyl, are prepared by either (i) reacting a compound of the Formula I wherein R 4 is hydrogen with a carboxyalkanoyl, alkanoyl, benzoyl or alkyl-substituted benzoyl halide or anhydride in the presence of an acid acceptor to give a compound of the invention wherein one of R 1 and R 3 is a group of the formula CH 2 OR 4 and R 4 is carboxyalkanoyl, alkanoyl, benzoyl or alkyl-substituted benzoyl, and optionally converting the compound wherein R 4 is carboxyalkanoyl to the ammonium salt thereof by reaction with ammonia, or (ii) reacting a compound of Formula I wherein one of R 1 and R 3 is formyl with an organolithium or an organomagnesium halide corresponding to R 6 , and hydrolysing the resulting organo metallic compound to give a compound of the invention wherein one of R 1 and R 3 is a group of the Formula II wherein R 5 is hydrogen and R 6 is as defined above. |
