Method for determination of normalised serum thyroxin levels comprises (i) extracting thyrosin (I) from thyroxin-binding globulin (TBG) by mixing he sample with an extracting agent which contains a dil. acid chosen from HCl; H2SO4; H3PO4; HNO3; HNO2; aliphatic amino-, carboxylic- or dicarboxylic acids; aromatic amino-, hydroxy- or dicarboxylic acids or their mixts., (II) mixing extracted (I) with TBG saturated with radiolabelled (I), the amt. being sufficient for labelling; (III) contacting the mixt. with a second binding agent (II), (IV) incubating, (V) measuring initial radioactivity of the incubated mixt., (VI) separating the mixt. and measuring residual activity in remaining (II).. Since acid extraction destroys the (I)-TBG complex, the method gives a value of (I) concn. compensated for changes in TGB levels (which may occur during pregnancy, oestrogen therapy etc). So gives a true indication of the status of the thyroid gland. |
