<p>Glucose is determined in an aq. sample by (a) diluting the sample in an aq. diluent buffered to pH 4-8 (pref. 5-7), (b) passing the dil. sample through a bed of glucose oxidase held on a solid carrier, with a residence time sufficient to oxidise the glucose, with formation of H2O2, (c) removing the mixt. from the bed and passing it into direct contact with a system of polarographic electrodes, (d) determining the H2O2 polarographically by determn. of the electric current, which is directly proportional to the H2O2 concn., and (e) calculating the glucose concn. from this current. Claimed apparatus consists of a reaction chamber, with sample inlet and prod. outlet, and contg. the bed of supported enzyme, and a polarographic cell attached to the prod. outlet. The cell consists of a tubular chamber, with inlet and outlet, contg. a system of polarographic electrodes which are in direct contact with the sample; the electrodes are connected to an ampere meter for determn. of the current developed in the cell.</p> |
