<p>Cpds. of formula : and their acid-addn. salts (where n is 1,2 or 3; R1 is OH; R2 is H, alkyl; or R1 with R2 is =O; R3, R4 = H, cycloalkyl or R3+R4 and the attached N-atom form a heterocyclic ring (opt. substd.)). In mice doses of 2-10 mg/kg. induce sedation. Higher doses abolish spontaneous motor activity and muscular hypotonicity. There is no loss of righting reflex, even at doses of 100-500 mg/kg. The tranquillising ED50 in mice ranges from 4-16 mg/kg orally. Clinical dosage units contain 5-200 mg. of active component.</p> |
