<p>New antibiotic A-3459A ("chloquinomycin") has characteristics which include C 56.25%, H 3.64%, O 32.07%, Cl 6.63%, N 0%; molecular weight 614 (vapour phase osmometry; m.pt. 228-238 degrees C (dec.); alpha 20D=+15 degrees (c=0.01, CHCl3) readily soluble in dimethylformamide, dimethyl sulphoxide, pyridine and aqueous NaOH; soluble in acetone, ethyl acetate, CHCl3, benzene, Et2O, acetic acid, MeOH, EtOH and BuOH; insoluble in n-hexane, petroleum ether and water. The acetate of antibiotic A-3459A (C 59.5%, H 4.25%, N 0%; m.pt. 188-200 degrees C) is also claimed. A-3459A and its acetate have antibiotic activity against gram-positive organisms, including antibiotic-resistant strains. MIC's of A-3459A and its acetate are given e.g. A3459A against Staph. aureus 209P, B. subtilis PCI 219 and E. coli they are 0.8, 0.4 and >50 mcg/ml, respectively. A-3459A is produced by cultivation of Streptomyces californicus var. rivalis ATCC 31173 and is converted into its acetate by reacting with acetic anhydride in pyridine. Cultivation conditions comprise aerobic conditions, ca 20-38 degrees C and pH ca 5.5-8.5 in a nutrient medium contg. usual carbon and nitrogen sources and inorganic salts.</p> |
