PURPOSE: To separate the lymphocyte and monocyte in a whole blood sample accurately by filling a container with a thixotropic gel and a chemical reagent and disposing a membrane for preventing absorption of water from the whole blood sample or the like between them. CONSTITUTION: A container 12 is filled, at the bottom part thereof, with a water soluble thixotropic gel 14 chemically inactive to the components of blood and filled, at the upper part thereof, with a chemical reagent 28 through a hydrophobic barrier 32. The chemical reagent 28 reacts on a whole blood sample filling the upper space 26 thereof to cause variation in the diameter and density of granulocyte and communicates with the thixotropic gel 14. The hydrophobic barrier 32 prevents absorption of water by the thixotropic gel 14 from the chemical reagent 28 and/or unseparated whole blood sample before lymphocyte and monocyte are separated from each other. According to the arrangement, water absorption has no effect on the cell separation characteristics of thixotropic gel 14. |
