<p>1483222 Viral complex; vaccines LABORATOIRE ROGER BELLON 3 Sept 1974 [4 Sept 1973] 38383/74 Heading A5B [Also in Divisions C3 an C6] A viral complex composed of viral particles -bivalent cations-ethylene oxide polymer, which may be used for the concentration and purification of viruses and in the production of vaccines, is prepared by admixing with a suspension of viral particles, virulent or attenuated, an ethylene oxide polymer and a bivalent metal cation which together form an insoluble complex on to which the viral particles are absorbed. The bivalent metal cation, preferably calcium (or magnesium), may be contained in the polymer used. Additional amounts of the cation may, however, be introduced into the suspension e.g. as CaCl 2 . Preferably, the ethylene oxide polymers are of M.W. 100000- 300000 and they are used in proporitions ranging from 2 to 4% weight/volume of suspension. The method of concentrating and purifying viral suspensions involves dissolving the viral complex in the required quantity of a solution containing a chelating agent for the bivalent metal cation to obtain a viral suspension having a higher concentration of viral particles than that from which the insoluble viral complex was produced and separating off any insoluble material. By repeating the steps in say, three cycles, viral subparticles and antigens may be eliminated and purified viral suspensions concentrated by the order of 1500 with respect to the original suspension may be obtained, Allergising substances may be precipitated out by treating the suspension with chloroform. The chelating agent may be EDTA or citric acid (or salts thereof). The method of preparing a vaccine may involve suspending the viral complex in the minimum quantity of a buffer solution at pH 7.6 containing bivalent metal cations to produce a paste, suspending the paste in a physiological buffer solution and preparing a vaccine from the suspension thus obtained by conventional procedures. The examples are directed to the concentrating and purifying of aphthous fever, influenza, Newcastle disease, rabies and poliomyelitis viruses of specified type (or origin) and mode of cultivation. In the procedure of Examples 13, 17 and 19 a purified and concentrated suspension of aphthous fever or rabies virus is compounded with alumina and saponin. The viral preparation may be attenuated by the addition of glycidaldehyde or: formaldehyde. Example 20 discloses a mixed antiaphthous fever/anti-rabies vaccine.</p> |
