<P>PROBLEM TO BE SOLVED: To provide a PCR (polymerase chain reaction) primer set for detection/determination of nitrous acid oxidoreductase gene, nor B, which a nitrous acid oxidative bacterium has. <P>SOLUTION: A primer is prepared based on a base sequence of the nitrous acid oxidoreductase gene which Nitrobacter hamburgensis has. PCR is carried out by using DNA of activated sludge as a template by using primer sets 1 and 2 composed of two kinds of primers, and PCR conditions which are optimal to each primer are obtained. After the PCR product is refined, the refined PCR product is linked to a plasmid vector and Escherichia coli is transformed to provide a clone. As a result of blast search, each clone obtained from the primer sets 1 and 2 exhibits high homology with nor B gene of Nitrobacter hamburgensis. Quantitative determination of numbers of copy of nor B gene is also made possible by using C-PCR method. <P>COPYRIGHT: (C)2005,JPO&NCIPI |
