PURPOSE:To advantageously produce and obtain the subject protease having improved heat stability by having disulfide bondings in a molecule by transforming a host with a recombinant plasmid transduced with a gene coding a specific alkali protease of modified Aspergillus oryzae and then culturing. CONSTITUTION:At least, two amino acids in alkali protease of wild Aspergillus oryzae are respectively substituted with cystine to prepare a gene coding alkali protease of modified Aspergillus oryzae having disulfide bondings in a molecule. Next, said gene is transduced into a plasmid to prepare a recombinant plasmid. Then, a host is transformed by using said recombinant plasmid. Thus, the resultant transformant is cultured in a medium and above-mentioned alkali protease of modified Aspergillus oryzae is produced, collected by separation to afford the objective alkali protease of modified Aspergillus oryzae useful in a field of food, etc., having excellent heat resistance. |
