PURPOSE:To prevent the escape of a culturing object and to enable the culture of plant tissue in high density by vertically moving a culture liquid surface with a specific means within a range to keep the stationary state of the culturing object on a baffle plate in a culture tank. CONSTITUTION:Culture liquid 10 in a culture tank 1 is extracted with a pump 3 and introduced to a tank 2. The pump is stopped when the level of the culture liquid 10 is higher than the uppermost baffle plate 9a having a number of perforations on the whole surface of a part of the surface. The size of the perforation is selected to inhibit the passage of the callus or hair root of the plant. A electromagnetic valve 8 is opened simultaneously with the stop of the pump and the culture liquid 10 extracted from the culture tank 1 is again returned to a culture tank 11. During the pulsation stirring to periodically or intermittently move the surface of the culture liquid 10 in vertical direction, the culturing object keeps its stationary state without moving in vertical direction. The culture of the tissue can be carried out in high proliferation rate by this process to improve the efficiency of the biosynthetic production of a useful substance by tissue culture.
