A polypeptide-containing fat-mobilising substance, FMS-1A, and a polypeptide-containing anorexigenic substance, FMS-1B, are prepared from the urine of fasting or cold-acclimatised animals by addition, at an acidic pH, of an alkanol to give a precipitate, FMS-1, comprising the mixed crude substances, and extracting the precipitate firstly with water to give a solution of FMS-1A and then with dilute aqueous alkali to give a solution of FMS-1B. The word "anorexigenic" is said to mean capable of causing loss of appetite. The animals, preferably rats, or horses, goats, sheep, dogs, cows, or rabbits, may be fasted by first feeding on a high fat diet for several days, followed by removal of all food, with urine collection over the next 24 hours; the cold acclimatisation is effected by transfer of the animal kept at room temperature, to an environment at 5 DEG C. for 24 hours to 3 days, with concomitant urine collection. FMS-1 is precipitated with, e.g. methanol, ethanol or propanol, from the urine acidified to pH 5.3 with benzoic, acetic, hydrochloric or sulphuric acid. The alkali may be aqueous alkali-metal or alkaline earth metal hydroxide or carbonate. The two substances FMS-1A and 1B may be purified and obtained in solid form by re-precipitation from the solutions with acidified alcohol, with subsequent dissolution of the precipitate in water or dilute alkali (whichever is appropriate), and lyophilisation. Also described is the preparation of a similar polypeptide, "Hypox FMS", by the same process but from urine from fasted hypophysectomised rats. This substance has anorexigenic activity, but no effect upon fat metabolism. FMS-1A and 1B are also said to cause hypoglycemia. The invention includes injectable compositions of FMS-1A and 1B with an inert carrier. |
