A method for detecting single-stranded DNA in a sample is new and comprises contacting the sample with a forward and/or reverse primer (I), at least one of which carries at least part of a label system in a 3' mismatched portion of the primer. The mismatched region is at least one, preferably at least 2-5 nucleotides long. At least one template-dependent polymerase with a 3'-5' proofreading activity cleaves the annealed labeled primer in its mismatched region to release the label and this release is then detected. An Independent claim is included for a kit for the detection of a target nucleic acid in a sample, comprising (I), and at least one nucleic acid polymerase.
