PRODUCTION OF HUMAN TYPE CHIMERA ANTIBODY

摘要

PURPOSE:To provide the method for producing the human type chimera antibody entirely without changing the amino acid sequence of a mouse antibody variable region. CONSTITUTION:A cDNA coding the heavy chain variable region of the antibody of an animal excluding man and a cDNA coding the light chain variable region of the antibody of the animal excluding the man are inserted into a cassette vector containing a cDNA coding the heavy chain constant region of the human antibody and into a cassette vector containing a cDNA coding the light chain constant region of the human antibody, respectively, with a synthetic DNA comprising the 5'-terminal side base sequence of the constant region of the antibody of the man and the 3'-terminal side base sequence of the variable region of the antibody of the animal excluding the man. The recombined cassette vectors are introduced into an animal cultured cell, which is cultured in a medium to produce and accumulate the human type chimera antibody in the cultured product. The human type chimera antibody is collected from the cultured product.
