It is intended to provide a method of conveniently detecting a gene mutation, detecting a restriction enzyme-recognition sequence, or detecting or quantifying the amplification of a specific sequence by <i>in vitro</i> reactions in a homogeneous assay system, namely, without resort to any complicated procedure such as electrophoresis or so-called Bound/Free separation; and an assay kit to be used in this detection method. A probe containing a signal generating agent is hybridized with a target nucleic acid and then treated with an endonuclease and an exonuclease or a protein having an exonuclease activity to thereby release the signal generating agent. Thus, a gene mutation, a restriction enzyme-recognition sequence or a specific sequence can be detected at a high sensitivity, thereby achieving the above object. |
