Purified hepatitis B antigen - obtd. by elimination of beta-lipoprotein, dissociation of complex and denaturing protein contaminants

Prepn. of purified HBs antigen from a serum prepn. comprises conventional methods and, in addn., (a) elimination of beta-lipoproteins, (b) dissociation of the immune complex contg. HBs into an insol. phase with elimination of the contaminant proteins and (c) denaturing without rendering insoluble, the residual contaminant proteins. This provides removal of complexes with immunoglobulins which reduce the concn. of anti-HBs antibody and reduce the immunogenic activity of the prepn. The prepn. may also be more likely to produce hypersensitivity reactions.
