A combination biosynthetic method of 5-O-desosaminyl erythronolide A comprises the steps of: (i) producing a glycosyltransferase gene eryBV adhering an L-mycarose ugar chain by acting on Saccharopolyspora erythraea erythronolide B; and (ii) cultivating a strain which additionally generates a substrate-thixotropic cytochrome P450 monooxygenase heterologous gene pikC in an Streptomyces venezuelae-derived pikromycin biosynthetic pathway, to Saccharopolyspora erythraea eryBV-deletion mutant and extracting the 5-O-desosaminyl erythronolide A from the culture fluid.
