PURPOSE: To discriminate a leukocyte accurately by treating a whole blood with an active composition for fixing the leukocyte and mixing a fluorochrome mixture, stimulating the fluorochrome at some wavelength and then measuring the relative intensity of fluorescence emission for at least two wavelength ranges. CONSTITUTION: An active composition (pretreating solution) for dissolving and fixing (containing lower aldehyde and Tris buffer) and a coloring reagent (containing at least one kind of coloring matter selected, respectively, from thiazole group and aminoxanthene group) are prepared. 50μl of blood is placed in a test tube, for example, and 50μl of a pretreating reagent is dripped by means of a pipet. The solution is incubated for 5-120sec while stirring and added with 500μl of a buffer followed by addition of 100μl of coloring reagent solution while oscillating. Subsequently, the solution is incubated for 5-120sec and fed to a flow cytometer. The encapsulated stream is composed of a buffer having pH in the range of 7-8. A light having wavelength of 515nm is employed for irradiation and an emitted fluorescence is measured over the wavelength of 500-550nm and 570-630nm.
