摘要 PURPOSE:To obtain a desired polypeptide in high productivity and easy separation and purification by producing a desired polypeptide having relatively low molecular weight as a fused protein with a specific penicilase in subjecting the desired peptide to in vitro secretion production using Escherichia coli. CONSTITUTION:DNA coding a desired polypeptide having relatively low molecular weight is coupled to DNA coding penicillinase derived from an alkalophilic Bacillus through DNA coding amino acid sequence specifically recognized by proteinasae having high specificity such as urokinase or DNA coding amino acid sequence specifically recognized by chemical substance such as cyanogen bromide. A microorganism is transformed by a vector in which the resultant coupled DNA is integrated and a fused protein is produced by the transformed microorganism. The resultant fused protein is separated by urokinase or cyanogen bromide to provide the desired polypeptide.
