PURPOSE: Provided are prenatal diagnostic reagent and kit for preterm birth, fetal infection and fetal damage by determining the concentration of matrix metalloproteinase-8(MMP-8) of the mother. The diagnostic kit is less stressful, and more sensitive and specific than the conventional method for determining cytokine from the blood of fetus. The reagent and the kit can not only diagnose symptoms of early rupture of fetal membranes and preterm birth but also diagnose preterm birth, fetal infection and fetal damage from the asymptomatic mother. CONSTITUTION: The diagnosis method of preterm birth, fetal infection and fetal damage comprises the steps of sampling amniotic fluid and measuring the concentration of MMP-8(matrix metalloproteinase) in the sampled amniotic fluid. The measurement method of MMP-8 in amniotic fluid comprises the steps of: adsorbing MMP-8 antigene, which is either monoclonal or multiclonal, to substrate; washing the substrate after adding amniotic fluid to the above substrate and reacting it; adding and reacting antigene bound with fluorescent material with the above substrate; and adding coupler to the above substrate and induce coloring for the observation of the specificity of the antigene reaction. The diagnostic kit includes MMP-8 antigene, standard MMP-8, substrate, analytical buffer solution, secondary antigene marked with coloring enzyme or fluorescent material and sealing plate cover.
