PREPARATION OF COLONIZATION STIMULATION FACTOR AND KALLIKREIN ORIGINATED FROM HUMAN URINE

PURPOSE:To separate and purify CSF (colonization stimulation factor) and kallikrein which are physiologically active substances, in high recovery, by the high- speed liquid gel-filtration chromatography of the urine of healthy man or a solution containing CSF and kallikrein. CONSTITUTION:Urine of healthy man or a solution containing CSF and kallikrein originated from human urine is concentrated in terms of proteins, and the concentrated liquid is made to equilibrate with a buffer solution containing a stabilizer (e.g. polyethylene glycol having a molecular weight of 1,000-10,000 Dalton) [e.g. a buffer solution having an ionic strength of 0.05-0.5 and pH of 6.0-8.0 and containing 0.01-0.1% (W/V) of the stabilizer]. The equilibrated mixture is passed through a column packed with a carrier having a molecular sieve effect having a molecular weight exclusion limit of 10<5>-5X10<5> Dalton measured by globular protein and treated preliminarily with said buffer solution. CSF and kallikrein are separated and recovered from the liquid by the high-speed liquid gel-filtration chromatography.
