PROBLEM TO BE SOLVED: To provide a method of manufacturing a room temperature-hardening organopolysiloxane composition which has appropriate thixotropy before hardening and gives a highly hard hardened product and a coating film after hardening. SOLUTION: In the method of manufacturing the above-mentioned composition containing an organopolysiloxane I which is a condensation product of an organopolysiloxane A containing M and Q units at a molar ratio (M/Q) of 0.6-1.2, having optionally D and T units respectively in 1.0 mole or less per 1 mole of the Q unit, less than 2.0 mass% of SiOH group, and an organopolysiloxane B terminal-sealed with a functional group-containing silyl group; a silane compound II having two or more silicon atom-bonded hydrolytic groups and/or its partial hydrolytic condensate; a filler III; a catalyst IV and a solvent V; a mixture is obtained by mixing B with III, the mixture is mixed with the solvent V of A, A and B are condensed in the presence of IV to obtain I-containing mixture, and the mixture is mixed with II to obtain the composition.
