The invention relates to a method for the production of nucleoside-diphosphate sugar (NDP-sugar), in which one sugar-1-phosphate reacts with a nucleoside-triphosophate in the presence of a bivalent metal cation as cofactor via a pyrophosphorylase that transforms sugar-1-phosphate. In accordance with the invention, the NTP concentration or preferably the concentration ratio between NDP-sugar and the bivalent metal cation is kept constant. Surprisingly, during such a reaction, inhibition of the pyrophosphorylase is greatly prevented, thanks to which the reaction, on the basis of constant enzyme activity, demonstrates high space-time-yield.
