Retroviral vectors are provided to improve safety of use by minimizing activation of genes around the target gene, and maintain high titer of virus and high expression rate of target gene, so that the retroviral vectors are useful for gene therapy. The MLV(murine leukemia virus)-derived recombinant retroviral vector is prepared by deleting the nucleotide number x to y in the U3 sequences of LTR(long terminal repeat) for MLV, wherein x is 7800-7900 and y is 8100-8260; the structural genes of MLV including gag, pol and env genes are completely deleted; and the recombinant retroviral vector further contains a heterologous gene such as enhanced green fluorescence gene or gp91-phox gene and an operatively linked heterologous promoter such as IE(immediate-early) promoter of human cytomegalovirus. A recombinant plasmid pI-LND-n contains (1) the MLV-derived recombinant retroviral vector, and (2) an expression cassette containing a selection marker gene, an operatively linked heterologous promoter and a polyadenylation signal.
