PURPOSE: To obtain the subject gene useful for increasing secretion ability of esterase carrying out hydrolysis reaction from a bacterium. CONSTITUTION: This gene encodes a polypeptide participating to an esterase secretion mechanism of a bacterium belonging to the genus Serratia, has a base sequence of the formula and is a DNA having three open reading frames(ORF). A cell of a bacterium such as Serratia marcescens Sr 41 (FERM BP-487) is treated with lysozyme and a surfactant such as sodium laurate, extracted with an organic solvent such as phenol and precipitated with ethanal to give a chromosomal DNA, which can be amplified by a conventional method. |
