发明名称 MODIFIED HYDROGENASE AND ENZYMATIC ELECTRODE USING THE SAME, AND METHOD FOR MODIFYING HYDROGENASE
摘要 <P>PROBLEM TO BE SOLVED: To provide a modified hydrogenase which is high in resistance to oxygen and excellent in stability, to provide an enzymatic electrode which uses the modified hydrogenase high in the oxygen resistance and exhibits electrode characteristics stable over a long period, and to provide a method for modifying a hydrogenase to obtain the modified hydrogenase having high oxygen resistance. <P>SOLUTION: This modified hydrogenase is prepared by removing an electron-transmitting site of an electron-transmitting subunit from a hydrogenase comprising (1) an active subunit containing an active site having a hydrogen-oxidizing or reducing activity and (2) the electron-transmitting subunit containing the electron-transmitting site for delivering electrons between the active site and the outside of the hydrogenase. The modified hydrogenase is obtained by exposing a hydrogenase to an oxygen atmosphere, when the hydrogenase is separated from a microbe producing the hydrogenase and (1) the active subunit containing the active site having the hydrogen-oxidizing or reducing activity. The enzymatic electrode is provided by using the modified hydrogenase. And the method for modifying the hydrogenase comprises a process for separating a hydrogenase from a microbe containing (1) the active subunit containing the active site having the hydrogen-oxidizing or reducing activity and (2) the electron-transmitting subunit containing the electron-transmitting site for delivering electrons between the active site and the outside of the hydrogenase, and the like, to remove the electron-transmitting site of the electron-transmitting subunit from the hydrogenase. <P>COPYRIGHT: (C)2009,JPO&INPIT
