An isolated and purified TACE polypeptide that inhibits the TNF-á proteolytic activity is useful in the manufacture of a medicament for treating conditions such as inhibiting the cleavage of TNF-á from cell membranes in a mammal, blocking the binding TNF-á with TACE in a mammal, disease characterized by an overproduction or an up-regulated production of TNF-á such as systemic inflammatory response syndrome, reperfusion injury, cardiovascular disease, HIV infection, HIV neuropathy, obstetrical or gynecological disorders, inflammatory disease, allergic disease and such other disease described herein, wherein the TACE polypeptide comprises an amino acid sequence selected from the group described in the specification. |
