摘要 Column chromatography apparatus comprising a chromatography column 270 comprising an inlet and an outlet. The inlet comprises an end fitting 280 through which mobile phase is introduced to column 270 in at least two separate portions which are independently controllable, and in different radial regions of the column. The inlet may comprise a frit assembly 278 configured to segment the mobile phase into at least two separate portions. Preferably the inlet comprises a flow distributor. Preferably, eluate leaves the column through a single outlet, and preferably the outlet splits the eluate into at least two separate portions which may be processed separately. A first portion of eluate may emanate from a central radial region of the column. A method of column chromatography comprises: providing to the inlet of a chromatography column at least two separate portions of mobile phase which are independently controllable, at least one of which contains a sample; introducing the at least two separate portions of mobile phase into different radial regions of the column; and flowing the portions of mobile phase longitudinally through the column in the different radial regions to effect separation of the sample into components.
