PROBLEM TO BE SOLVED: To provide a new RNA transcription method using as a template a modified nucleic acid(DNA) with very high exo- and endo-nuclease tolerance.

SOLUTION: The new RNA transcription and/or amplification method is characterized by involving using as the template the modified DNA where hydroxy group(s) and/or oxygen atom(s) bound to phosphorus atom(s) of at least one phosphoric acid moiety is(are) substituted with other atom(s) and/or group(s).
