The present invention relates to a method for improving the drug metabolism function of a human stem cell-derived hepatocyte. Although the human stem cell-derived hepatocyte is similar to a human hepatocyte in shape, it was confirmed that expression of drug or alcohol metabolism-related enzymes and antioxidant enzymes has been reduced. Hereupon, as a result of co-culturing a human stem cell-derived hepatocyte with a mouse hepatic stellate cell, it was confirmed that hepatocyte damage and the degree of cell death were reduced due to a decrease in toxicity caused by alcohol, and expression of drug and alcohol metabolism-related enzymes and antioxidant enzymes has been increased. In addition, as a result of co-culturing a human stem cell-derived hepatocyte with a mouse hepatic stellate cell, retinol-related gene expression was changed. Also, as a result of co-culturing, with a human stem cell-derived hepatocyte, a hepatic stellate cell isolated from a mouse which does not metabolize retinol into retinoic acid, hepatocyte damage was not reduced, and expression of drug and alcohol metabolism-related enzymes and antioxidant enzymes was not changed. Thus, as a result of treating a human stem cell-derived hepatocyte with retinoic acid, it was confirmed that expression of drug and alcohol metabolism-related enzymes and antioxidant enzymes increased, and the degree of cell death decreased. Accordingly, the method for co-culturing a human stem cell-derived hepatocyte with a hepatic stellate cell, and treating the human stem cell-derived hepatocyte with retinoic acid can be useful as a method for improving the drug metabolism function of the human stem cell-derived hepatocyte. |
