METHOD OF DETERMINING BASE SEQUENCE OF DNA

It is intended to provide a method of determining the base sequence of a DNA. In this method of determining the base sequence of a DNA, use is made of a probe which has a protruding end, has been labeled for identification depending on the kind of the base at the protruding end and contains the recognition sequence of class IIS restriction enzyme. For a number of DNAs to be analyzed, the linkage of the terminal base of each DNA to be analyzed to the probe and the cleavage of the terminal base of the DNA to be analyzed are simultaneously conducted in chain reactions. Then, the base sequence can be successively determined by the single-molecule fluorometry. Thus, the base sequences of these DNAs can be efficiently determined.
