PROBLEM TO BE SOLVED: To provide a spectrofluorometric analysis method of a single-molecule, capable of precisely measuring an interaction of molecules. SOLUTION: The single-molecule fluorometric analyzing technique is to use fluorometric resonance energy transfer (FRET). That is, the spectrofluorometric analyzing method is adopted, in order to detect the association or interaction of a sample A and a sample B and includes a process for bringing the sample labeled with fluorescence into contact with the sample B labeled with a fluorescent substance, which converts the fluorescence energy of the fluorescent substance for use in labeling the sample A to exciting energy; a process of emitting light, having the wavelength for exciting the fluorescent substance for use in labeling the fluorescent substance to irradiate the fluorescent substance; a process for detecting the fluorescence emitted from the fluorescent substance, used for labeling the samples A and B; and a process for performing a fluorescence correlation spectral method or fluorescence intensity distribution analysis on the basis of the detected fluorescences to detect the change in the reaction of the samples A and B.
