PROBLEM TO BE SOLVED: To provide monovalent antibodies having a long half life when administered in vivo, preparation methods of such monovalent antibodies, pharmaceutical compositions comprising such antibodies, and use of such monovalent antibodies.SOLUTION: The invention provides: a nucleic acid construct encoding a light chain of a monovalent antibody comprising a nucleotide sequence encoding the VL region of an antigen-specific antibody functionally linked to a nucleotide sequence encoding a constant CL region of an Ig, where the nucleic acid construct is modified in such a manner that the CL region does not contain amino acid(s) that may form a disulfide bond or a covalent bond with other peptides comprising the same amino acid sequence of the CL region when administered to an animal or a human; and a nucleic acid construct encoding a heavy chain of a monovalent antibody comprising a nucleotide sequence encoding the VH region of an antigen-specific antibody functionally linked to a nucleotide sequence encoding a constant CH region of a human Ig, where the nucleic acid construct is modified in such a manner that the heavy chain does not contain amino acid(s) that may form a stable non covalent bond with other heavy chains when administered to an animal or a human.SELECTED DRAWING: None |
